In 1968, Dharmsinh Desai, who was then a member of Parliament, established the Dharmsinh Desai Institute of Technology as a centre for higher education in the fields of engineering and technology. In 1991, it received autonomy under Gujarat University, and in June 2000, it received Deemed University status.

The institute was renamed Dharmsinh Desai University in 2005 after achieving full university status. In 2005, the university began offering a bachelor's degree in dental surgery, and in 2006, the university opened its Faculty of Pharmacy. It is the first university in Gujarat to receive approval to begin offering degrees in nanotechnology in 2012. The Medical Council of India has given the institution permission to open a new medical college in the 2019–20 academic year.
